Lead compound
The most promising compound found to bind a drug target, chosen as the starting point that medicinal chemists will modify. A lead usually has useful activity but also flaws, such as low potency, side effects or poor absorption.
Example
Penicillin, discovered in a mould in 1928, was the lead for a whole family of modified antibiotics such as amoxicillin.
